THC Y CBD INHIBEN LA PROLIFERACIÓN DE CÉLULAS CANCERÍGENAS EN LOS PULMONES

Los pacientes con cáncer de pulmón de células no pequeñas (CPCNP)
desarrollan resistencia a los agentes antitumorales por mecanismos que
involucran la transición epitelial a mesoesenquimatosa (EMT).
Esto requiere el desarrollo de nuevos medicamentos complementarios agonistas de los receptores de cannabinoides (CB1 y CB2), incluidos el tetrahidrocannabinol (THC) y el cannabidiol (CBD).
Se evaluó la relación entre los niveles de expresión del CB1 y el CB2
con las características clínicas de pacientes con CPCNP, y
el efecto del THC y el CBD (individualmente y en combinación) sobre la
proliferación, la EMT y la migración in vitro en las líneas celulares de
cáncer de pulmón A549, H460 y H1792.
Los cambios morfológicos en las células se visualizaron por contraste de
fase y microscopía de fluorescencia. La migración se estudió mediante
la recolonización por rascado inducida por el factor de crecimiento
epidérmico (EGF) de 20 ng/ml.
Las muestras del tumor se clasificaron según el nivel de expresión de
CB1, CB2, o ambos. Los pacientes con altos niveles de expresión de CB1,
CB2 y CB1/CB2 mostraron un aumento de la supervivencia que alcanzó la
importancia para CB1 y CB1/CB2 (p = 0,035 y 0,025, respectivamente).
Ambos agonistas cannabinoides inhibieron la proliferación y expresión
del EGFR en las células de cáncer de pulmón, y el CBD potenció el efecto
del THC.
Por su lado, el CBD y el THC por sí solos o en combinación, restauraron el fenotipo epitelial.
Por último, ambos cannabinoides redujeron la migración in vitro de las líneas utilizadas.
